In Arabidopsis, GLUT1 (AtGLUT1) and GLUT9 (AtGLUT9) are two members of the major facilitator superfamily. Mutations in AtGLUT1 prevent the import of glucose into the cell, causing growth retardation and sterility. Loss-of-function alleles of AtGLUT9 cause a severe phenotype in sugar-rich conditions, including increased susceptibility to bacterial infection, collapse of the root meristem, and reduced seedling viability. To examine how AtGLUT1 and AtGLUT9 function in Arabidopsis, we generated double and triple mutants by crossing and analyzed the growth phenotype of transgenic plants expressing Cre recombinase from the natural AtGLUT1 and AtGLUT9 promoters. Here, we report that the double mutants are viable on medium with high sugar concentration and that the mutant phenotype is partially suppressed by mutations in AtGLUT1, but not in AtGLUT9, suggesting that AtGLUT9 may be partially redundant with AtGLUT1. We also demonstrate that a putative lignin precursor in Arabidopsis, caffeic acid 3-O-methyltransferase (AtC3MT), constitutively accumulates in the double mutants, indicating that lignification may be impaired in the double mutants.
